It needs to be installed, selected _and_ opened.

- To open it, double click on the actual dictionary file either in the finder or in the catalog.

- To check if it is open (i.e. in use), set the catalog window to the folder containing the definition dictionary and check to see that the icon is dimmed.

- Once the dictionary is opened, the spell check dialog should add a "?" button right below the checkmark button, and a scrollable window should appear at the bottom on the left. The "?" button and scrollable window will be dimmed unless there is something in the "Replace with:" box.

But be forewarned: the definition dictionary is known to cause crashes for some people.

And finally one other point: as far as I know the definition dictionary is only available in U.S. English. I'm not sure if I recall correctly, but I remember some discussion about incompatability between the two sets. Certainly my attempts today at mixing dictionaries from different languages (in the preferences) were not successful.
